Description

This mouse monoclonal antibody, supplied by STEMCELL Technologies, is clone 16A and targets human MUC1 (CD227). MUC1 is a large (>250 kDa), heavily glycosylated type 1 transmembrane protein expressed on the surface of most glandular and ductal epithelial cells and on a variety of hematopoietic cells. The clone 16A antibody has a higher affinity for the glycosylated form of MUC1, and the target is also known by alternative names including CD227, EMA, Episialin, Epithelial membrane antigen, HMFG antigen, MAM6, Mucin 1, PEM, and Polymorphic epithelial mucin.

The reagent is conjugated with PE (phycoerythrin) and was raised in C57BL/6 mice. It is of the IgG1, lambda isotype and was generated using Jurkat cells expressing MUC1 as the immunogen. The antibody was purified by affinity chromatography and conjugated with PE under optimal conditions; the solution is free of unconjugated PE and unconjugated antibody. It is formulated as a phosphate-buffered solution at pH 7.2, containing 0.09% sodium azide and 0.2% (w/v) bovine serum albumin (BSA), and is supplied as a clear liquid.

Flow cytometry (FC) is the verified and reported application for this clone, which has been used to quantify airway apical epithelial cells cultured in PneumaCult-ALI Medium at the air-liquid interface. For flow cytometry, the suggested use is 5 μL per 1 x 10^6 cells in 100 μL, with titration recommended for optimal performance in each application. The product is stable at 2 - 8°C when stored undiluted; it should not be frozen and should be protected from prolonged exposure to light. According to the safety data sheet, the mixture is not classified as hazardous under the 2012 OSHA Hazard Communication Standard or the GHS.
