Anti-Hu MUC1 (CD227), 16A, AZ, 25tests

STEMCELL Technologies offers this mouse monoclonal antibody specific for human MUC1 (also known as CD227), supplied as clone 16A conjugated to the APC fluorophore. The target is a large (> 250 kDa) heavily glycosylated type 1 transmembrane protein expressed on the surface of most glandular and ductal epithelial cells as well as on a variety of hematopoietic cells. Its extracellular portion can extend beyond most cell-surface proteins, suggesting a role in cell-cell and cell-substrate adhesion, and it is highly expressed by a majority of human adenocarcinomas where it is associated with a poor prognosis. The 16A clone has a higher affinity for the glycosylated form of MUC1.

The antibody is produced in C57BL/6 mice and is of the IgG1, lambda isotype, raised against Jurkat cells expressing MUC1. It has been verified for flow cytometry and is suited to quantifying airway apical epithelial cells cultured in PneumaCult-ALI Medium at the air-liquid interface. The suggested working amount for flow cytometry is 5 μL per 1 x 10^6 cells in 100 μL, although titration is recommended for optimal performance.

The product is formulated in a phosphate-buffered solution, pH 7.2, containing 0.09% sodium azide and 0.2% (w/v) BSA. It is purified by affinity chromatography and conjugated with APC under optimal conditions, and the solution is free of unconjugated APC and unconjugated antibody. The antibody should be stored undiluted at 2 - 8 degrees C; it must not be frozen and should be protected from prolonged exposure to light.
