Description

A rabbit polyclonal antibody from STEMCELL Technologies that reacts with E-cadherin (CD324), a transmembrane glycoprotein located in the adherens junction of epithelial cells. E-cadherin is a calcium-dependent cell-cell adhesion protein composed of a large extracellular domain with five cadherin-motif subdomains, a single-pass transmembrane segment, and a short conserved cytoplasmic domain. It serves as a key regulator of epithelial junction formation, associating with catenins and linking to cortical actin bundles at both the zonula adherens and the lateral adhesion plaques; disruption of this complex by tyrosine phosphorylation alters cell adhesion properties. The antigen plays a central role in tissue architecture and integrity, and its expression has been implicated in tumor metastasis.

The antibody was raised in rabbits against recombinant human E-cadherin protein (Met1 - Ile707) and is unconjugated. It has been verified for ICC/IF and reported for use in IHC-P and WB. Suggested starting concentrations are ICC/IF at 2.5 μg/mL, IHC-P at 1:500 - 1:2000, and WB at 1:500 - 1:2000, with titration recommended for optimal performance in each application. The reagent has been verified for labeling E-cadherin-positive intestinal epithelial cells in human intestinal organoids grown using the STEMdiff™ Intestinal Organoid Kit. Alternative antigen names include Arc-1, CD324, CDH1, CDHE, E-cad, E-Cadherin, ECAD, LCAM, and UVO, with Gene ID 999.

The antibody is supplied in phosphate-buffered saline and purified by affinity chromatography. For short-term storage, keep the product at 2 - 8°C for up to one month when stored undiluted; for longer-term storage, aliquot and store at -20°C, and do not re-freeze thawed aliquots. A third-party listing additionally recommends protecting from light and avoiding repeated freeze–thaw cycles.
