Description

This mouse monoclonal IgG1, kappa antibody from STEMCELL Technologies, Clone SK1, is conjugated with PE (phycoerythrin) and binds to CD8a, a 32–34 kDa type I transmembrane glycoprotein that forms a subunit of the CD8 co-receptor. The antibody recognises CD8a on cells from human, rhesus, cynomolgus, chimpanzee, African green monkey, pigtailed macaque, and sooty mangabey samples, and is raised in BALB/c mice using human peripheral blood T lymphocytes as the immunogen.

CD8 is expressed either as an αβ heterodimer or an αα homodimer and acts as a co-receptor with the T cell receptor during MHC class I-restricted antigen recognition, strengthening the association between the TCR and the MHC I–antigen complex and amplifying intracellular signalling through interaction with cytoplasmic tyrosine kinases such as Lck. Reportedly, the SK1 antibody exhibits a blocking effect on MHC:peptide tetramer binding to the TCR and also blocks proliferation of Leu-2 T cells in mixed leukocyte reactions.

Application use has been verified for flow cytometry (FC). The suggested use for flow cytometry is 5 µL per 1 × 10^6 cells in 100 µL or per 100 µL of whole blood, and titration is recommended for optimal performance. The reagent is supplied as a phosphate-buffered solution at pH 7.2 containing 0.09% sodium azide and 0.2% (w/v) bovine serum albumin, and the antibody was purified by affinity chromatography prior to PE conjugation. The product is stable at 2–8 °C when stored undiluted; it should not be frozen and should be protected from prolonged exposure to light.
