Anti-Hu CD8a, RPA-T8, FI, 25tests

From STEMCELL Technologies, this mouse monoclonal IgG1, kappa antibody (clone RPA-T8) is conjugated to FITC and recognizes human CD8a, a 32–34 kDa type I transmembrane glycoprotein that serves as a subunit of the CD8 co-receptor. CD8 binds MHC Class I molecules during T cell activation, strengthening the association between the T cell receptor and the MHC–antigen complex. The antibody labels CD8a expressed by a majority of thymocytes, a subset of mature peripheral blood T cells (cytotoxic T cells), a proportion of γδ T cells, and at lower levels on NK cells.

Reactivity has been demonstrated with human, rhesus, cynomolgus, baboon, chimpanzee, pigtailed macaque, and sooty mangabey samples. The antibody was raised against full-length human CD8 protein and purified by affinity chromatography, with the conjugate confirmed free of unconjugated FITC. It is supplied in a phosphate-buffered solution at pH 7.2, containing 0.09% sodium azide and 0.2% (w/v) bovine serum albumin. For flow cytometry, the manufacturer recommends 5 µL per 1 x 10^6 cells in 100 µL or per 100 µL of whole blood, with titration advised for optimal performance.

The reagent is suitable for flow cytometry and cell isolation, and is stable at 2–8°C when stored undiluted. It should not be frozen and should be protected from prolonged exposure to light.
