Description

A PE-conjugated monoclonal antibody from STEMCELL Technologies, clone HB15e, specifically targets human CD83 and is intended for immunophenotyping by flow cytometry [S2][S3]. The reagent is of mouse IgG1, kappa isotype, and the HB15e clone is reported to bind an epitope on CD83 that overlaps with that of antibody 1E11 [S3].

CD83 is an approximately 45 kDa type I transmembrane glycoprotein belonging to the Siglec protein family, containing an extracellular V-type Ig-like domain and a C-terminal cytoplasmic tail; a soluble form of CD83 has also been identified [S3]. The molecule is expressed predominantly on circulating and tissue dendritic cells, including Langerhans cells and interdigitating reticulum cells, with weaker expression reported on activated lymphocytes, and may also be found on neutrophils, monocytes, and macrophages [S1][S3]. CD83 expression is typically transient, except on dendritic cells, and correlates with upregulation of HLA class II antigen [S3].

Beyond flow cytometry, the antibody is also described as suitable for immunoblotting and immunohistochemistry applications, with reported reactivity against baboon, chimpanzee, cynomolgus monkey, human, rhesus monkey, and macaque samples [S3]. CD83 is described as functionally important for the development and maintenance of the immune system, with suggested roles in cell adhesion and signaling, antigen presentation, and lymphocyte activation; the soluble form of CD83 appears to be immunosuppressive [S3]. For reagent handling, the conjugate should be stored at 2–8 °C, protected from light, with repeated freeze–thaw cycles avoided, and is intended for research use only [S2].
