Anti-Hu CD83, HB15e, PE, 100µg

A PE-conjugated monoclonal antibody from STEMCELL Technologies, clone HB15e, recognizes human CD83 (Gene ID 9308) — an approximately 45 kDa type I transmembrane glycoprotein and member of the Siglec (sialic acid-binding immunoglobulin-like lectin) family. The reagent is a mouse IgG1, kappa antibody raised against human CD83-transfected African green monkey COS-7 cells, purified by affinity chromatography and conjugated with PE under optimal conditions; the solution is free of unconjugated PE and unconjugated antibody.

Species reactivity has been characterized on human, rhesus, cynomolgus, baboon, chimpanzee, and pigtailed macaque samples. Verified and reported applications are limited to flow cytometry. For flow cytometry, the suggested use is ≤ 5 μL (test size products) or ≤ 2 μg (μg size products) per 1 × 10^6 cells in 100 μL volume or per 100 μL of whole blood, and titration is recommended for optimal performance. The antibody clone has also been verified for purity assessments of cells isolated with EasySep™ kits and for analyzing human monocyte-derived mature dendritic cells.

CD83 is expressed predominantly on circulating and tissue dendritic cells, including Langerhan cells and interdigitating reticulum cells, with weak expression on activated lymphocytes and additional reported expression on neutrophils, monocytes, and macrophages; CD83 expression serves as a marker for dendritic cell maturation and activation. A soluble form of CD83 has also been identified and reportedly appears to be immunosuppressive, blocking T cell activation and inhibiting dendritic cell maturation. The product is formulated in a phosphate-buffered solution at pH 7.2, containing 0.09% sodium azide, may contain carrier protein/stabilizer, and should be stored undiluted at 2 - 8°C, protected from prolonged exposure to light and not frozen.
