Anti-Hu CD83, HB15e, FI, 100tests

Manufactured by STEMCELL Technologies, this mouse monoclonal antibody (clone HB15e, isotype IgG1, kappa) is conjugated to FITC and targets CD83, an ~45 kDa type I transmembrane glycoprotein and member of the Siglec (sialic acid-binding immunoglobulin-like lectin) protein family. The antibody was generated using human CD83-transfected African green monkey COS-7 cells as the immunogen and reacts with CD83 on human, rhesus, cynomolgus, baboon, chimpanzee, and pigtailed macaque samples (Gene ID: 9308). The antigen is also known as B-cell activation protein, BL11, and HB15, contains an extracellular V-type Ig-like domain and a C-terminal cytoplasmic tail, and has a soluble form that has also been identified.

The reagent binds CD83 expressed predominantly on circulating and tissue dendritic cells, including Langerhans cells and interdigitating reticulum cells, as well as weakly on activated lymphocytes, neutrophils, monocytes, and macrophages. CD83 expression correlates with upregulation of HLA class II antigen and serves as a marker for dendritic cell maturation and activation. The HB15e epitope reportedly overlaps with that of antibody 1E11. The conjugate has been evaluated by flow cytometry and verified for purity assessments of cells isolated with EasySep™ kits and for analyzing human monocyte-derived mature dendritic cells cultured with StemSpan™-ACF medium. For flow cytometry, a suggested use of ≤ 5 μL per 1 x 10^6 cells in 100 μL volume is provided, and titration for optimal performance is recommended.

The antibody is supplied in phosphate-buffered solution, pH 7.2, containing 0.09% sodium azide and 0.2% (w/v) bovine serum albumin, and was purified by affinity chromatography before conjugation; the solution is free of unconjugated FITC. The product should be stored at 2 - 8 °C undiluted, protected from prolonged exposure to light, and not frozen. For research use only.
