Anti-Hu CD83, HB15e, AZ, 100tests

This laboratory reagent from STEMCELL Technologies is a mouse monoclonal antibody (Clone HB15e, IgG1, kappa) that reacts with human CD83, an approximately 45 kDa type I transmembrane glycoprotein and member of the Siglec protein family containing an extracellular V-type Ig-like domain and a C-terminal cytoplasmic tail. The antibody is conjugated to APC (allophycocyanin).

CD83 is expressed predominantly on circulating and tissue dendritic cells, including Langerhans cells and interdigitating reticulum cells, with weaker expression reported on activated lymphocytes and presence on neutrophils, monocytes, and macrophages. Antigen expression is typically transient, except on dendritic cells, and correlates with upregulation of HLA class II. While CD83 function is described as ill-defined, evidence suggests it is essential for the development and maintenance of the immune system, with proposed roles in cell adhesion and signaling, antigen presentation, and lymphocyte activation. A soluble form of CD83 has also been identified and is reported to be immunosuppressive. The epitope bound by Clone HB15e reportedly overlaps with that of antibody 1E11.

Validated applications include flow cytometry, Western blot, and immunohistochemistry. Flow cytometry use has been demonstrated on dendritic cells derived from human peripheral blood mononuclear cells (PBMCs).
