Anti-Hu CD83, HB15e, AD, 100tests

STEMCELL Technologies supplies this Alexa Fluor® 488-conjugated mouse monoclonal antibody, clone HB15e (IgG1, kappa), raised against human CD83-transfected African green monkey COS-7 cells. The reagent shows reactivity against human, rhesus, cynomolgus, baboon, chimpanzee, and pigtailed macaque samples (target antigen Gene ID: 9308).

CD83 is an ~45 kDa type I transmembrane glycoprotein of the Siglec (sialic acid-binding immunoglobulin-like lectin) family, featuring an extracellular V-type Ig-like domain and a C-terminal cytoplasmic tail; a soluble form has also been identified. CD83 is expressed predominantly on circulating and tissue dendritic cells, including Langerhan cells and interdigitating reticulum cells, weakly on activated lymphocytes, and may also be found on neutrophils, monocytes, and macrophages. Its expression is typically transient except on dendritic cells, correlates with upregulation of HLA class II antigen, and serves as a marker for dendritic cell maturation and activation. Although its function is ill-defined, CD83 appears essential for immune system development and is implicated in cell adhesion and signaling, antigen presentation, and lymphocyte activation and proliferation. The soluble form appears immunosuppressive, blocking T cell activation and inhibiting dendritic cell maturation. The epitope bound by HB15e reportedly overlaps with that of antibody 1E11.

Flow cytometry (FC) is both the verified and reported application. The suggested use is ≤ 5 μL per 1 x 10^6 cells in 100 μL volume or 5 μL per 100 μL of whole blood, with titration recommended for optimal performance. This clone has been used for purity assessments of cells isolated with EasySep™ kits, including the EasySep™ Human CD14 Positive Selection Kit, and for analyzing human monocyte-derived mature dendritic cells cultured using StemSpan™-ACF medium. The product is provided in phosphate-buffered solution, pH 7.2, containing 0.09% sodium azide and 0.2% (w/v) bovine serum albumin. It was purified by affinity chromatography and conjugated with Alexa Fluor® 488, with the solution free of unconjugated Alexa Fluor® 488. Store undiluted at 2 - 8 °C; do not freeze, and protect from prolonged exposure to light.
