Description

STEMCELL Technologies offers this PE-conjugated mouse monoclonal IgG1, kappa antibody, Clone AD2, raised against the human CD73 antigen (also known as 5'-nucleotidase, ecto; Ecto-5'-nucleotidase; L-VAP-2; NT5E). The clone was generated in BALB/c mice using the human pre-B leukemia cell line 207 as immunogen, and the antibody was purified by affinity chromatography before conjugation with PE; the solution is free of unconjugated PE and unconjugated antibody.

CD73 is a glycosyl phosphatidylinositol (GPI)-anchored glycoprotein and ecto-5'-nucleotidase expressed on the surface of subsets of B and T cells, follicular dendritic cells, mesenchymal stem cells, endothelial cells, and epithelial cells. It forms a homodimer of ~70 kDa subunits that contact each other through their C-terminal domains and catalyzes the hydrolysis of 5'-adenosine monophosphate (AMP) to form the bioactive nucleoside adenosine, playing a pivotal role in the activation of P1 adenosine receptors. CD73 also functions as a co-signaling molecule on T cells and as an adhesion molecule mediating lymphocyte interactions with the endothelium and follicular dendritic cells. It serves as a marker of lymphocyte differentiation and for identifying undifferentiated mesenchymal stem cells, and is highly expressed in many types of human and mouse cancers, where it has been implicated in tumor growth control.

The antibody cross-reacts with human, rhesus, chimpanzee, and pigtailed macaque samples, and is verified for flow cytometry, with reported use in FACS, IF, and IHC. It has been verified for purity assessments of cells isolated with EasySep kits and for use with MesenCult media for human mesenchymal cell labeling. The reagent is formulated in a phosphate-buffered solution, pH 7.2, containing 0.09% sodium azide and 0.2% (w/v) bovine serum albumin. The suggested use for flow cytometry is ≤ 5 μL per 1 x 10^6 cells in 100 μL, with titration recommended for optimal performance. The product is stable when stored undiluted at 2 - 8°C; do not freeze and protect from prolonged exposure to light.
