Description

This PE-conjugated monoclonal antibody from STEMCELL Technologies targets human CD73, a glycosyl phosphatidylinositol (GPI)-anchored glycoprotein that functions as ecto-5'-nucleotidase. Clone AD2, raised in BALB/c mice, is of the IgG1, kappa isotype and was generated using a human pre-B leukemia cell line (207) as immunogen. The antibody reacts with CD73 on human, rhesus, chimpanzee, and pigtailed macaque samples, and is verified for flow cytometry, with reported use in FACS, immunofluorescence microscopy, and immunohistochemistry.

CD73 is expressed on subsets of B and T cells, follicular dendritic cells, mesenchymal stem cells, endothelial cells, and epithelial cells. The molecule forms a homodimer of ~70 kDa subunits and catalyzes the hydrolysis of 5'-adenosine monophosphate (AMP) to adenosine, regulating extracellular adenosine concentrations and P1 adenosine receptor signaling. According to the manufacturer, this clone has been applied for purity assessments of cells isolated with EasySep™ kits and for labeling human mesenchymal cells grown in MesenCult™ media, and serves as a marker for identifying undifferentiated mesenchymal stem cells.

The reagent is supplied in a phosphate-buffered solution (pH 7.2) containing 0.09% sodium azide and 0.2% (w/v) bovine serum albumin, and has been purified by affinity chromatography prior to conjugation with PE. For flow cytometry, the suggested use is ≤ 5 μL per 1 x 10^6 cells in 100 μL. The product should be stored at 2 - 8°C, protected from prolonged exposure to light, and not frozen. This reagent is supplied by STEMCELL Technologies for research use only.
