Anti-Hu CD73, AD2, FI, 25tests

This FITC-conjugated monoclonal antibody from STEMCELL Technologies targets CD73 (Ecto-5'-Nucleotidase), a glycosyl phosphatidylinositol (GPI)-anchored glycoprotein expressed on the surface of subsets of B and T cells, follicular dendritic cells, mesenchymal stem cells, endothelial cells, and epithelial cells. Produced in Mouse (BALB/c) as a monoclonal IgG1, kappa antibody, clone AD2 was raised against the human pre-B leukemia cell line 207 and reacts with CD73 from human, rhesus, chimpanzee, and pigtailed macaque samples. The target antigen forms a homodimer of ~70 kDa subunits and functions as an ecto-5'-nucleotidase that catalyzes the hydrolysis of 5'-adenosine monophosphate (AMP) to form the bioactive nucleoside adenosine, thereby regulating extracellular adenosine concentrations and activating P1 adenosine receptors. CD73 also acts as a co-signaling molecule on T cells and as an adhesion molecule mediating lymphocyte interactions with the endothelium and follicular dendritic cells, and is used as a marker for lymphocyte differentiation and for identifying undifferentiated mesenchymal stem cells.

The antibody has been verified for flow cytometry (FC) and reported for FC and immunofluorescence, and has been verified for purity assessments of cells isolated with EasySep™ kits and for labeling human mesenchymal cells grown in MesenCult™ media. It is supplied as a clear liquid in phosphate-buffered solution, pH 7.2, containing 0.09% sodium azide and 0.2% (w/v) bovine serum albumin, and is purified by affinity chromatography then conjugated with FITC; the solution is free of unconjugated FITC and unconjugated antibody. For flow cytometry, the suggested use is ≤ 5 μL per 1 x 10^6 cells in 100 μL, with titration recommended for each application. The product is stable at 2 - 8°C when stored undiluted; do not freeze and protect from prolonged exposure to light. According to the supplier's safety data sheet, the product is not classified as a hazardous substance or mixture under GHS or the 2012 OSHA Hazard Communication Standard, and STEMCELL Technologies' quality management system is certified to ISO 13485.
