Anti-Hu CD73, AD2, AZ, 100tests

This mouse monoclonal antibody from STEMCELL Technologies targets human CD73 (also known as ecto-5'-nucleotidase, 5'-nucleotidase, L-VAP-2, and NT5E; Gene ID 4907) and is provided as clone AD2 conjugated to the APC fluorochrome.

CD73 is a glycosyl phosphatidylinositol (GPI)-anchored glycoprotein that functions as a homodimer of ~70 kDa subunits and acts as an ecto-5'-nucleotidase, catalyzing the hydrolysis of 5'-adenosine monophosphate (AMP) to the bioactive nucleoside adenosine. It is expressed on subsets of B and T cells, follicular dendritic cells, mesenchymal stem cells, endothelial cells, and epithelial cells, and is implicated in P1 adenosine receptor activation, T cell co-signaling, and lymphocyte adhesion to endothelium and follicular dendritic cells. CD73 is used as a marker of lymphocyte differentiation, with expression increasing during development, and is also used to identify undifferentiated mesenchymal stem cells. It is reported to be highly expressed in many human and mouse cancers and has been linked to several immunodeficiency diseases when genetically defective.

The IgG1, kappa antibody was raised against the human pre-B leukemia cell line 207 in BALB/c mice and purified by affinity chromatography prior to conjugation with APC. It is formulated in a phosphate-buffered solution at pH 7.2 containing 0.09% sodium azide and 0.2% (w/v) bovine serum albumin. Applications verified by flow cytometry (FC) are listed, with reported use in immunofluorescence microscopy (IF) and immunohistochemistry (IHC). Reactivity is established for human, rhesus, chimpanzee, and pigtailed macaque samples. For flow cytometry, the suggested use is ≤ 5 µL per 1 x 10^6 cells in 100 µL; titration for optimal performance is recommended. The product is stable at 2 - 8°C when stored undiluted, should be protected from prolonged exposure to light, and must not be frozen.
