Description
From STEMCELL Technologies, this Anti-Human CD71 (Transferrin Receptor) Antibody, Clone OKT9, FITC is a mouse monoclonal IgG1, kappa antibody directed against human CD71 (transferrin receptor). The OKT9 antibody reacts with CD71, described as an approximately 180–190 kDa disulfide-bonded homodimer and type 2 transmembrane glycoprotein expressed at high levels on a broad range of actively proliferating cells, as well as on reticulocytes, monocytes, macrophages and marrow stromal cells. Surface levels are low on resting leukocytes but are upregulated on lymphocytes, monocytes and macrophages following antigen or mitogen stimulation, and expression is lost during differentiation of reticulocytes into mature erythrocytes.
CD71 mediates uptake of transferrin–iron complexes through receptor-mediated endocytosis and recycling of the apotransferrin–receptor complex to the cell surface, with two molecules of iron-loaded transferrin bound by the receptor and expression regulated by metabolic demand for iron. CD71 has been employed as a marker for evaluating erythroid precursors within the bone marrow and for assessing disorders such as erythroid leukemia and myelodysplastic syndrome, and it has also served as a target for drug delivery. The OKT9 antibody binds an epitope that reportedly overlaps with that of antibody clone CY1G4 but is distinct from the ligand binding site for transferrin. The antibody was generated using human acute lymphoblastic leukaemia cells as the immunogen (Gene ID 7037).
For flow cytometry, the suggested use is ≤ 5 μL (0.125 μg) per 1 × 10⁶ cells in 100 μL volume, with titration recommended for optimal performance for each application. The antibody has been verified for flow cytometry, and this clone has been verified for purity assessments of cells isolated with EasySep kits, including the EasySep Direct Human T Cell Isolation Kit and EasySep Human CD3 Positive Selection Kit, as well as for peripheral blood erythroid precursor cells cultured with StemSpan SFEM II medium and StemSpan Erythroid Expansion Supplement. The product is supplied in aqueous buffer containing 0.09% sodium azide, was purified by affinity chromatography, and is free of unconjugated FITC. It is stable at 2–8 °C when stored undiluted; the solution should not be frozen and should be protected from prolonged exposure to light. The product is intended for research use only.
