Anti-Hu CD71, OKT9, AZ, 25tests

From STEMCELL Technologies, the Anti-Human CD71 (Transferrin Receptor) Antibody, Clone OKT9, is a mouse monoclonal IgG1, kappa antibody conjugated with APC (Allophycocyanin) and verified for flow cytometry. The antibody targets CD71, also known by alternative names including TFRC, TFR1, and TRFR (Gene ID: 7037), an ~180–190 kDa disulfide-bonded homodimer and type 2 transmembrane glycoprotein that mediates uptake of transferrin–iron complexes through receptor-mediated endocytosis. It was generated using human acute lymphoblastic leukemia cells as the immunogen.

CD71 is expressed at high levels on a broad range of actively proliferating cells, as well as on reticulocytes, monocytes, macrophages, and marrow stromal cells. Surface levels are low on resting leukocytes but are upregulated on lymphocytes, monocytes, and macrophages following antigen or mitogen stimulation, while expression is lost during differentiation of reticulocytes into mature erythrocytes. The OKT9 antibody binds an epitope that reportedly overlaps with that of antibody clone CY1G4 but is distinct from the ligand-binding site for transferrin. CD71 has been employed as a marker for evaluating erythroid precursors within the bone marrow and for assessing disorders such as erythroid leukemia and myelodysplastic syndrome, and it has also served as a target for drug delivery.

The antibody is supplied in a phosphate-buffered solution, pH 7.2, containing 0.09% sodium azide, 0.1% gelatin, and 0.2% (w/v) bovine serum albumin, and has been purified by affinity chromatography. For flow cytometry, the suggested use is ≤ 5 µL (0.06 µg) per 1 x 10^6 cells in 100 µL, with titration recommended for optimal performance. The product is stable at 2–8°C when stored undiluted and should not be frozen or exposed to prolonged light. This antibody clone has been verified for purity assessments of cells isolated with EasySep™ kits, as well as for peripheral blood erythroid precursor cells cultured with StemSpan™ SFEM II medium.
