Anti-Hu CD68, Y1/82A, FI, 100tests

From STEMCELL Technologies, this FITC-conjugated mouse monoclonal antibody of the IgG2b, kappa isotype, clone Y1/82A, targets human CD68 — an approximately 110 kDa type 1 transmembrane glycoprotein belonging to the sialomucin, LAMP and scavenger receptor families. The Y1/82A clone recognizes an epitope distinct from those of clones Y2/131, EBM11, Ki-M6 and KP1, and is reportedly more specific for monocytes and macrophages than KP1.

CD68 is highly expressed in lysosomes, endosomes and cytoplasmic granules, and more weakly on the surface of macrophages, monocytes, neutrophils, basophils, dendritic cells and NK cells. It has also been detected in the cytoplasm of γ/δ T cells, LAK cells, fibroblasts, endothelial cells and subsets of B cells and hematopoietic progenitors, as well as in non-hematopoietic tissues such as liver and kidney. The antigen is widely used as a marker for cells of the macrophage lineage, for example in distinguishing monocyte/macrophage from lymphoid forms of leukemia.

The antibody was raised against phytohaemagglutinin-activated peripheral blood mononuclear cells in BALB/c mice, purified by affinity chromatography and conjugated with FITC under optimal conditions, with the solution free of unconjugated FITC. It is formulated in a phosphate-buffered solution at pH 7.2, containing 0.09% sodium azide and 0.2% (w/v) bovine serum albumin. Flow cytometry (FC) is both a verified and reported application; the clone has been validated for purity assessments of cells isolated with EasySep kits, including the EasySep Human CD14 Positive Selection Kit, and for analyzing human macrophages derived from cultured monocytes. Suggested usage is ≤ 5 μL per 1 × 10^6 cells in 100 μL volume, or 5 μL per 100 μL of whole blood, with titration recommended for optimal performance.
