Description

A mouse monoclonal antibody from STEMCELL Technologies, clone Y1/82A, recognizes human CD68, an ~110 kDa type I transmembrane glycoprotein belonging to the sialomucin, LAMP, and scavenger receptor families. Its heavily glycosylated extracellular domain contains a proline-rich spacer region separating mucin and LAMP domains, and the molecule has been shown to bind low-density lipoprotein along with certain lectins and selectins, although its precise function remains unclear.

CD68 is highly expressed in lysosomes, endosomes, and cytoplasmic granules, with weaker surface expression on macrophages, monocytes, neutrophils, basophils, dendritic cells, and NK cells. It has also been detected in the cytoplasm of γ/δ T cells, LAK cells, fibroblasts, endothelial cells, subsets of B cells, and hematopoietic progenitors, as well as in non-hematopoietic tissues including liver and kidney. Clone Y1/82A recognizes an epitope distinct from those of clones Y2/131, EBM11, Ki-M6, and KP1, and is reportedly more specific for monocytes and macrophages than KP1.

This APC-conjugated reagent has an IgG2b, kappa isotype and is tested for flow cytometry, with the broader antibody format also validated for immunocytochemistry, immunofluorescence, immunohistochemistry, immunoprecipitation, and Western blot. According to the manufacturer's safety data sheet, the preparation is a clear liquid classified as not hazardous under GHS and intended for use as a laboratory reagent.
