Description

A FITC-conjugated monoclonal antibody from STEMCELL Technologies, Clone G10F5, reacts with human and chimpanzee CD66b, an ~95 kDa glycosylphosphatidylinositol (GPI)-linked glycoprotein expressed by neutrophilic and eosinophilic granulocytes. CD66b belongs to the carcinoembryonic antigen (CEA)-like subfamily of the immunoglobulin superfamily and possesses two Ig-like C2-type domains and one Ig-like V-type domain.

The antibody is of mouse origin with an IgM, kappa isotype, raised against human granulocytes. In neutrophils, CD66b is localised within cytoplasmic granules and on the plasma membrane, with surface expression levels increasing upon granulocyte activation by stimuli such as phorbol myristate acetate (PMA), calcium ionophore and N-formylmethionyl-leucyl-phenylalanine (FMLP). It acts as a signalling receptor involved in cell adhesion, phagocytosis and chemotaxis, and interacts with partners including CD66c, galectin-3, fibronectin and E-selectin.

Verified for flow cytometry (FC), the reagent is supplied in a phosphate-buffered solution at pH 7.2 containing 0.09% sodium azide and 0.2% (w/v) bovine serum albumin, with >85% purity and free of unconjugated FITC. The suggested use is 5 μL per 1 × 10^6 cells in 100 μL volume, or per 100 μL of whole blood. Product should be stored at 2–8°C, protected from prolonged light exposure and not frozen.
