Description
This FITC-conjugated monoclonal antibody from STEMCELL Technologies, clone G10F5, is of mouse IgM kappa isotype and specifically binds human CD66b. CD66b (previously designated CD67) is an approximately 95 kDa single-chain glycosylphosphatidylinositol (GPI)-linked glycoprotein expressed by neutrophilic and eosinophilic granulocytes. It belongs to the carcinoembryonic antigen (CEA)-like subfamily within the immunoglobulin superfamily and possesses two Ig-like C2-type domains and one Ig-like V-type domain. The antibody is also reported to react with chimpanzee cells.
In neutrophils, CD66b is localized within cytoplasmic granules and on the plasma membrane, and surface expression increases upon granulocyte activation by stimuli such as phorbol myristate acetate (PMA), calcium ionophore, and N-formylmethionyl-leucyl-phenylalanine (FMLP). CD66b functions as a signaling receptor involved in cell adhesion, phagocytosis, and chemotaxis, interacting with CD66c, galectin-3, fibronectin, and E-selectin. Clone G10F5 is verified for flow cytometry, with demonstrated labeling of human whole blood nucleated cells, buffy coat nucleated cells, and peripheral blood mononuclear cells (PBMCs), including following isolation with EasySep™ kits. The reagent should be stored at 2–8 °C, shielded from light, with repeated freeze–thaw cycles avoided. It is intended for research use only.
