Anti-Hu CD66b, G10F5, AZ, 25tests

Manufactured by STEMCELL Technologies, this monoclonal antibody (clone G10F5) targets CD66b, a glycosylphosphatidylinositol (GPI)-linked glycoprotein expressed on neutrophilic and eosinophilic granulocytes. CD66b, previously known as CD67, is approximately 95 kDa and belongs to the carcinoembryonic antigen (CEA)-like subfamily of the immunoglobulin superfamily, possessing two Ig-like C2-type domains and one Ig-like V-type domain. In neutrophils, the antigen localizes to cytoplasmic granules as well as the plasma membrane, and surface expression increases upon activation by stimuli such as phorbol myristate acetate (PMA), calcium ionophore, and N-formylmethionyl-leucyl-phenylalanine (FMLP). CD66b functions as a signaling receptor involved in cell adhesion, phagocytosis, and chemotaxis; reported binding partners include CD66c, galectin-3, fibronectin, and E-selectin.

The antibody is of the IgM, kappa isotype and is verified for flow cytometry. Validation data demonstrate use across unconjugated, APC, FITC, PE, and Pacific Blue™ conjugations on human whole blood nucleated cells, buffy coat nucleated cells, and peripheral blood mononuclear cells, including in workflows paired with EasySep™ cell isolation kits. Reported reactivity includes human and chimpanzee samples, with additional applications including functional assays, immunocytochemistry, immunofluorescence, immunohistochemistry, immunoprecipitation, and Western blotting.
