Anti-Hu CD63, H5C6, UC,100µg

A mouse monoclonal IgG1, kappa antibody from STEMCELL Technologies (Clone H5C6) recognizes an extracellular epitope on human CD63, a 30–60 kDa type III lysosomal glycoprotein and member of the tetraspanin family. Tetraspanins contain four transmembrane domains, two extracellular loops, and short cytoplasmic N- and C-termini, and CD63 partners with integrins and other proteins to form tetraspanin-enriched microdomains in the plasma membrane. The antigen participates in cell activation, adhesion, differentiation, and tumor invasion, and is broadly expressed on monocytes, macrophages, activated basophils, fibroblasts, smooth muscle cells, and activated platelets. CD63 is also found in late endosomes, lysosomes, and secretory vesicles, and serves as a characteristic marker of extracellular vesicles (EVs); CD63 deficiency has been associated with Hermansky-Pudlak syndrome, a rare autosomal recessive disorder characterized by platelet dysfunction and lysosomal storage defects.

The antibody was raised in BALB/c mice against human adherent splenocytes and purified by affinity chromatography. It is supplied unconjugated in a phosphate-buffered solution at pH 7.2, with endotoxin levels measured at < 0.01 EU (< 0.001 ng) per μg of protein by Limulus amebocyte lysate (LAL) analysis. The product is stable at 2–8 °C when stored undiluted and should not be frozen.

Verified applications include flow cytometry (FC) and Western blotting (WB); reported applications include ELISA, FC, ICC, IF, IHC, immunoaffinity chromatography, IP, and WB. Suggested use is ≤ 1.0 μg per 1 × 10⁶ cells in 100 μL (or per 100 μL of whole blood) for FC and 1–2 μg/mL for WB. The clone has been verified for purity assessments of EVs isolated using EasySep™ Extracellular Vesicle Human Positive Selection Kits, and reactivity has been confirmed with human, rhesus, cynomolgus, chimpanzee, baboon, African green monkey, and capuchin monkey CD63.
