Description
This FITC-conjugated mouse monoclonal IgG2a, kappa antibody (clone NKI-M9) from STEMCELL Technologies recognizes human CD51, also known as integrin αV or vitronectin receptor α chain. CD51 is a type I transmembrane glycoprotein generated by post-translational cleavage into two subunits of 125 kDa and 24 kDa linked by a disulfide bond. It associates non-covalently with integrin β1 (CD29), β3 (CD61), β5, β6, or β8 to form heterodimeric cell adhesion receptors for extracellular matrix components such as fibrinogen, collagen, fibronectin, laminin, osteopontin, thrombospondin, vitronectin, and von Willebrand factor; the pairing of CD51 with CD61 forms the integrin αv/β3 receptor primarily involved in vitronectin binding. Beyond mediating adhesion and cytoskeletal organization, CD51-containing integrins participate in signal transduction and modulate cell proliferation, differentiation, and migration.
The antibody was generated using human melanoma cells as immunogen and is supplied in a phosphate-buffered solution at pH 7.2 containing 0.09% sodium azide and 0.2% (w/v) bovine serum albumin. It was purified by affinity chromatography and conjugated with FITC under optimal conditions, and the resulting solution is free of unconjugated FITC. Applications verified by the manufacturer include flow cytometry (FC), with reported use in FACS and FC; representative flow cytometry data are shown for human HT1080 fibrosarcoma cells. The suggested use for flow cytometry is 5 µL per 1 × 10^6 cells in 100 µL volume, or per 100 µL of whole blood, with titration recommended for optimal performance. The product is stable at 2–8°C when stored undiluted; it must not be frozen and should be protected from prolonged exposure to light.
CD51 is broadly expressed across many cell types, including endothelial cells, fibroblasts, monocytes, macrophages, platelets (at relatively low levels), and osteoclasts, and is also detected on hepatoma, melanoma, and neuroblastoma cell lines. The associated gene identifier is 3685.
