Description

A mouse monoclonal IgG2a, kappa antibody from STEMCELL Technologies targets human CD51, also known as integrin αV or the vitronectin receptor α chain. Clone NKI-M9 was raised against human melanoma cells and is purified by affinity chromatography in a phosphate-buffered solution at pH 7.2 containing 0.09% sodium azide. The unconjugated reagent has been verified for flow cytometry and is reported for ELISA, functional assay, and immunoprecipitation. According to the product sheet, the suggested use for flow cytometry is ≤ 2 µg per 1 × 10^6 cells in a 100 µL volume, with titration recommended for optimal performance.

CD51 is a type I transmembrane glycoprotein of approximately 125 kDa and 24 kDa subunits generated by post-translational cleavage and linked by a disulfide bond. It associates non-covalently with integrin β1 (CD29), β3 (CD61), β5, β6, or β8 to form heterodimeric receptors that bind extracellular matrix components including vitronectin, fibrinogen, fibronectin, collagen, laminin, osteopontin, thrombospondin, and von Willebrand factor. CD51 is expressed broadly on endothelial cells, fibroblasts, monocytes, macrophages, platelets (at relatively low levels), and osteoclasts, and is also found on hepatoma, melanoma, and neuroblastoma cells. The antibody has been verified for labeling human mesenchymal cells cultured in MesenCult™ Proliferation Kit (Human) and MesenCult™-XF Medium.

The product is stable when stored undiluted at 2–8 °C and should not be frozen. Clone NKI-M9 is reportedly not suitable for immunohistochemistry on paraffin-embedded sections.
