Anti-Hu CD5, UCHT2, PE, 25tests

This PE-conjugated antibody from STEMCELL Technologies is a mouse monoclonal IgG1, kappa reagent directed against human CD5 and produced using clone UCHT2. CD5 is an approximately 67 kDa single-chain type I glycoprotein and a member of the scavenger receptor superfamily. It is constitutively expressed on thymocytes, T cells, B cell subsets, peripheral blood dendritic cells, and some leukemia and lymphoma cells, including chronic B lymphocytic leukemia (B-CLL) cells, with low levels on thymocytes and high density on mature T cells.

CD5 modulates T and B cell receptor signaling, thymocyte maturation, and T cell/B cell interactions through its physical and functional associations with the T cell receptor/CD3 complex and the B-cell receptor, and studies with CD5 knockout mice indicate that CD5 negatively regulates antigen receptor-mediated signaling in thymocytes and mature T cells. The UCHT2 antibody binds to epitope 2 located in the extracellular domain of CD5, with putative ligands including CD5L and CD72. The immunogen was human thymocytes followed by Sézary T cells, and species reactivity includes Human, Rhesus, Cynomolgus, Capuchin Monkey, Chimpanzee, Common Marmoset, and Owl Monkey.

The antibody is verified for flow cytometry (FC) and for purity assessments of cells isolated with EasySep™ HLA kits. It is supplied as a clear liquid, phosphate-buffered at pH 7.2 with 0.09% sodium azide and 0.2% (w/v) bovine serum albumin, purified by affinity chromatography and conjugated with PE; the product is not classified as hazardous. For flow cytometry, the suggested use is 5 μL per 1 x 10^6 cells in 100 μL volume or per 100 μL of whole blood. The reagent is stable at 2 - 8°C when stored undiluted and should not be frozen; protect from prolonged exposure to light.
