Anti-Hu CD5, UCHT2, FI, 100tests

A FITC-conjugated mouse monoclonal antibody from STEMCELL Technologies is designed for the detection of human CD5 by flow cytometry. Clone UCHT2, also known by alternative designations including Leu1, Leu-1, Ly-1, Lymphocyte Antigen T1, T1, and Tp67, is an IgG1 kappa immunoglobulin generated in BALB/c mice using human thymocytes followed by Sézary T cells as the immunogen. The antibody was purified by affinity chromatography and conjugated with FITC under optimal conditions, with the solution free of unconjugated FITC.

CD5 is an ~67 kDa single-chain type I glycoprotein and a member of the scavenger receptor superfamily. It is constitutively expressed on thymocytes, T cells, B cell subsets, peripheral blood dendritic cells, and some leukemia and lymphoma cells, including chronic B lymphocytic leukemia (B-CLL) cells. CD5 is expressed at low levels on thymocytes and at high density on mature T cells, and putative ligands include CD5L and CD72. The UCHT2 antibody binds to epitope 2 located in the extracellular domain of CD5, with reactivity confirmed against human, rhesus, cynomolgus, capuchin monkey, chimpanzee, common marmoset, and owl monkey samples. Flow cytometry is the verified and reported application.

The reagent is formulated in a phosphate-buffered solution, pH 7.2, containing 0.09% sodium azide and 0.2% (w/v) bovine serum albumin. The product is stable at 2 - 8°C when stored undiluted; it should not be frozen and must be protected from prolonged exposure to light. For flow cytometry, the suggested use is 5 µL per 1 x 10^6 cells in 100 µL volume or per 100 µL of whole blood, with titration recommended for optimal performance in each application.
