Anti-Hu CD45RO, UCHL1, PS, 25tests

From STEMCELL Technologies, the Anti-Human CD45RO Antibody, Clone UCHL1, is a mouse (BALB/c) monoclonal IgG2a, kappa antibody conjugated to PerCP-Cy5.5. The UCHL1 clone reacts with an extracellular epitope on CD45RO, the shortest (~180 kDa) isoform of the CD45 transmembrane glycoprotein and a member of the protein tyrosine phosphatase family (receptor class 1/6 subfamily). CD45RO is expressed on activated and memory (but not naïve) T cells, some B cell subsets, activated monocytes and macrophages, and granulocytes, enhances both T cell receptor- and B cell receptor-mediated activation, and is a known ligand for CD22 on B cells.

The antibody was raised against a human IL-2-dependent T-cell line (CA1) and purified by affinity chromatography, with the final solution free of unconjugated PerCP-Cy5.5. Verified for flow cytometry, it is commonly paired with anti-CD45RA antibodies to discern memory and naïve T cells and has been used to identify T-cell lymphomas and leukemia. The reagent is supplied as a clear liquid in a phosphate-buffered solution at pH 7.2 containing 0.09% sodium azide. It remains stable when stored undiluted at 2 - 8 °C, should not be frozen, and should be protected from prolonged exposure to light.

For flow cytometry, the manufacturer's suggested use is ≤ 5 μL (0.25 μg) per 1 x 10^6 cells in a 100 μL volume, and titration is recommended for optimal performance in each application. Beyond its primary reactivity with human, chimpanzee, and common marmoset samples, the antibody reportedly cross-reacts to varying extents with cow, dog, mouse, rat, and some macaques (Pig-tailed, Rhesus).
