Anti-Hu CD45RO, UCHL1, PE, 25tests

A mouse (BALB/c) monoclonal IgG2a, kappa antibody from STEMCELL Technologies, clone UCHL1, recognizes an extracellular epitope on CD45RO — the shortest isoform of CD45. CD45RO is a type I transmembrane glycoprotein and a member of the protein tyrosine phosphatase receptor family. Alternative splicing of exons 4, 5, and 6 generates up to eight CD45 isoforms with molecular masses of 180 - 240 kDa; the ~180 kDa CD45RO isoform is expressed on activated and memory T cells (but not naïve T cells), some B cell subsets, activated monocytes and macrophages, and granulocytes. CD45RO enhances both T cell receptor- and B cell receptor-mediated activation and is a known ligand for CD22 on B cells. The UCHL1 clone is used to identify T cell lymphomas and leukemias and is commonly combined with anti-CD45RA antibodies to distinguish memory from naïve T cells, with the proportion of CD45RO+ memory T cells reportedly increasing with age.

The reagent is conjugated to PE (phycoerythrin) and has been verified for flow cytometry. It reacts with human, chimpanzee, and common marmoset samples, and reportedly cross-reacts to varying extents with mouse, rat, cow, dog, and some macaques (rhesus, pig-tailed). For flow cytometry, the suggested use is ≤ 5 μL per 1 x 10^6 cells in 100 μL, or per 100 μL of whole blood; titration is recommended for optimal performance for each application. The antibody has also been verified for purity assessments of cells isolated with EasySep™ memory T cell enrichment kits.

Provided as a clear liquid in phosphate-buffered solution, pH 7.2, containing 0.09% sodium azide and 0.2% (w/v) bovine serum albumin, the antibody is purified by affinity chromatography and conjugated with PE under optimal conditions; the solution is free of unconjugated PE and unconjugated antibody. The manufacturer's Safety Data Sheet states that the mixture is not considered hazardous under the 2012 OSHA Hazard Communication Standard (29 CFR 1910.1200). Store undiluted at 2 - 8°C; do not freeze and protect from prolonged exposure to light.
