Description

From STEMCELL Technologies, this mouse monoclonal IgG2a, kappa antibody (clone UCHL1) is conjugated to APC and targets human CD45RO, the shortest isoform of CD45. CD45RO is a ~180 kDa type I transmembrane glycoprotein and member of the protein tyrosine phosphatase family, generated by alternative splicing of the CD45 gene. It is expressed on activated and memory (but not naïve) T cells, some B cell subsets, activated monocytes, macrophages, and granulocytes, enhances T cell receptor- and B cell receptor-mediated activation, and serves as a known ligand for CD22 on B cells. The UCHL1 antibody has been used to identify T cell lymphomas and leukemia, commonly alongside anti-CD45RA reagents to distinguish memory from naïve T cells, with the proportion of CD45RO+ memory T cells typically increasing with age.

The reagent is supplied in an aqueous buffer containing 0.09% sodium azide and was purified by affinity chromatography before conjugation with APC under optimal conditions. For flow cytometry, the suggested use is ≤ 5 μL per 1 x 10^6 cells in 100 μL or per 100 μL of whole blood, with titration recommended for each application. The product is stable at 2 - 8°C when stored undiluted; it should not be frozen and should be protected from prolonged exposure to light. The UCHL1 epitope is destroyed by treatment with neuraminidase or O-glycosidase.

The antibody was raised in BALB/c mice against a human IL-2-dependent T cell line (CA1) and has been verified for flow cytometry. It has also been used to assess the purity of cells isolated with EasySep™ Human Memory CD4+ T Cell Enrichment and EasySep™ Human Memory CD8+ T Cell Enrichment Kits. Reported species reactivity covers human, chimpanzee, and common marmoset, with cross-reactivity of varying extent reported against mouse, rat, cow, dog, and some macaques (Rhesus, Pig-tailed).
