Description

STEMCELL Technologies offers a mouse monoclonal antibody (Clone UCHL1, IgG2a kappa) conjugated to allophycocyanin (APC) that recognizes CD45RO, the shortest isoform of the CD45 transmembrane protein tyrosine phosphatase. The clone was raised against a human IL-2-dependent T-cell line (CA1) in BALB/c mice and reacts with an extracellular epitope on CD45RO expressed on human, chimpanzee, and common marmoset cells; it reportedly cross-reacts to varying extents with cow, dog, mouse, rat, and some macaques (pig-tailed and rhesus).

The ~180-kDa CD45RO isoform is generated by alternative splicing that excises the exons encoding the RA, RB and RC extracellular regions, and is expressed on activated and memory (but not naïve) T cells, some B cell subsets, activated monocytes and macrophages, and granulocytes. CD45RO enhances both T-cell receptor- and B-cell receptor-mediated activation and serves as a ligand for CD22 on B cells. The UCHL1 epitope is destroyed by treatment with neuraminidase or O-glycosidase. This reagent has been employed to identify T-cell lymphomas and leukemia, and is commonly used in combination with antibodies against CD45RA to discern memory from naïve T cells; the proportion of CD45RO+ memory T cells typically increases with age.

The antibody has been verified for flow cytometry, with a suggested use of ≤ 5 μL per 1 x 10⁶ cells in 100 μL volume or per 100 μL of whole blood, and titration is recommended for optimal performance. It is supplied in an aqueous buffer containing 0.09% sodium azide, purified by affinity chromatography and conjugated with APC under optimal conditions; the solution is free of unconjugated APC and unconjugated antibody. Store the product undiluted at 2 - 8 °C; do not freeze, and protect from prolonged exposure to light.
