Anti-Hu CD45, HI30, PS, 25tests

A mouse monoclonal antibody from STEMCELL Technologies, the Anti-Human CD45 clone HI30 is conjugated to PerCP-Cy5.5 and binds all isoforms of CD45, a type I transmembrane glycoprotein expressed on the surface of most hematopoietic cells except mature erythrocytes, platelets and plasma cells. The clone also reacts with chimpanzee CD45 (Gene ID 5788) and has been verified for purity assessments of cells isolated with EasySep™ kits, including the Human T Cell Enrichment Kit, Human CD4+ T Cell Enrichment Kit, and Human CD45 Depletion Kit.

The antibody is supplied as a liquid in phosphate-buffered solution at pH 7.2, containing 0.09% sodium azide and 0.1% gelatin, and has been purified by affinity chromatography prior to conjugation; the final solution is free of unconjugated PerCP-Cy5.5. For flow cytometry, the suggested use is ≤ 5 μL (0.25 μg) per 1 x 10e6 cells in 100 μL volume, and titration for optimal performance in each application is recommended. The product is stable at 2 - 8°C when stored undiluted and should not be frozen or exposed to prolonged light.

Alternative names for the target antigen include B220, LCA, and T200. The clone is of the IgG1, kappa isotype and was raised against full-length human CD45 protein. It recognizes an extracellular epitope known as the Leucocyte Common Antigen, located proximal to the membrane and common to all CD45 isoforms. CD45 is a member of the protein tyrosine phosphatase family, with two tyrosine phosphatase domains in its large cytoplasmic portion that modulate intracellular substrates such as the Src kinases Lck and Fyn, and it functions in immunoregulatory processes including cell activation, growth, differentiation, and oncogenic transformation. Alternative splicing of exons 4, 5, and 6 gives rise to up to 8 isoforms with molecular masses in the range of 180 - 240 kDa.
