Anti-Hu CD45, HI30, PS, 100tests

A PerCP-Cy5.5-conjugated antibody from STEMCELL Technologies, this mouse monoclonal IgG1, kappa reagent is Clone HI30 and reacts with all isoforms of CD45 — a type I transmembrane glycoprotein expressed on the surface of most hematopoietic cells, except mature erythrocytes, platelets and plasma cells, with expression being lost during differentiation of those cell types. Reported species reactivity includes human and chimpanzee.

CD45 is a member of the protein tyrosine phosphatase family and participates in immunoregulatory processes such as cell activation, growth, differentiation and oncogenic transformation. Its large cytoplasmic portion contains two tyrosine phosphatase domains involved in modulating intracellular substrates including the Src kinases Lck and Fyn. Alternative splicing of three exons (4, 5, 6) encoding the extracellular RA, RB and RC polypeptide sequences gives rise to up to eight isoforms with molecular masses in the range of 180 - 240 kDa. The epitope recognised by Clone HI30 — the Leucocyte Common Antigen (LCA, T200, B220) — is an extracellular region proximal to the membrane and common to all isoforms.

The antibody was raised against full-length human CD45 protein and purified by affinity chromatography, then conjugated with PerCP-Cy5.5 under optimal conditions; the resulting solution is free of unconjugated PerCP-Cy5.5. It is supplied as a phosphate-buffered solution at pH 7.2 containing 0.09% sodium azide and 0.1% gelatin. Flow cytometry is a verified application. For flow cytometry, the suggested use is ≤ 5 μL (0.25 μg) per 1 x 10⁶ cells in 100 μL volume, and titration is recommended for optimal performance per application. The product is stable at 2 - 8 °C when stored undiluted; it should not be frozen and should be protected from prolonged exposure to light. Products are for research use only.
