Description

A biotin-conjugated mouse monoclonal antibody from STEMCELL Technologies, Clone HI30, is specific for human and chimpanzee CD45. The HI30 antibody recognizes all isoforms of CD45, a type I transmembrane glycoprotein expressed on the surface of most hematopoietic cells except mature erythrocytes, platelets, and plasma cells. CD45 belongs to the protein tyrosine phosphatase family and participates in immunoregulatory processes including cell activation, growth, differentiation, and oncogenic transformation. Its large cytoplasmic region contains two tyrosine phosphatase domains, one of which is enzymatically active, that modulate intracellular substrates such as the Src kinases Lck and Fyn.

The antibody was raised against full-length human CD45 protein and is of the IgG1, kappa isotype. Alternative splicing of three exons (4, 5, 6) encoding the extracellular RA, RB, and RC polypeptide sequences gives rise to up to 8 isoforms with molecular masses of 180 - 240 kDa. The leukocyte common antigen (LCA), the extracellular region proximal to the membrane that is recognized by HI30, is common to all isoforms of CD45.

Verified and reported applications are limited to flow cytometry (FC), with a suggested use of ≤ 0.125 µg per 1 x 10^6 cells in 100 µL. The antibody has been verified for purity assessments of cells isolated with EasySep™ kits. It is supplied as a clear liquid, purified by affinity chromatography and conjugated with biotin under optimal conditions; the solution is free of unconjugated biotin. The formulation is a phosphate-buffered solution, pH 7.2, containing 0.09% sodium azide. The product is stable at 2 - 8 °C when stored undiluted and should not be frozen.
