Description

STEMCELL Technologies offers a mouse monoclonal IgG1, kappa antibody (Clone 2D1) raised in BALB/c mice against human CD45 and conjugated to FITC. The clone reacts with all isoforms of CD45, a type I transmembrane glycoprotein expressed on the surface of hematopoietic stem and progenitor cells and on mature blood cells, but not on mature erythrocytes, platelets, or plasma cells; CD45 expression is lost as these lineages differentiate.

CD45 is a member of the protein tyrosine phosphatase family and is involved in immunoregulatory processes including cell activation, growth, differentiation, and oncogenic transformation. Its large cytoplasmic portion contains two tyrosine phosphatase domains that modulate intracellular substrates such as the Src kinases Lck and Fyn. Alternative splicing of three exons (4, 5, 6) generates up to 8 isoforms with molecular masses in the range of 180 - 240 kDa. The 2D1 antibody recognizes the Leukocyte Common Antigen, an extracellular region located proximal to the membrane and common to all isoforms of CD45.

The antibody is supplied in phosphate-buffered saline, pH 7.2, containing 0.09% sodium azide and 0.1% gelatin, and was purified by affinity chromatography before FITC conjugation; the solution is free of unconjugated FITC. The product has been verified for flow cytometry, with a suggested use of 5 μL per 1 x 10^6 cells in a 100 μL volume. It is stable at 2 - 8 °C when stored undiluted, should not be frozen, and should be protected from prolonged exposure to light.
