Anti-Hu CD43, CD43-10G7, PE, 25t

A PE-conjugated mouse monoclonal IgG1, kappa antibody supplied by STEMCELL Technologies targets human CD43, a type I transmembrane sialoglycoprotein expressed on the surface of most human hematopoietic cells. Clone CD43-10G7 was raised in BALB/c mice against the MV4-11 human myelomonocytic cell line and is verified for flow cytometry. According to the manufacturer, CD43 is found on thymocytes, peripheral T cells, NK cells, monocytes, granulocytes, activated B cells, an early B cell subset in bone marrow, plasma cells, and hematopoietic progenitor cells, with low expression on platelets.

CD43 displays different glycoforms in the range of ~95 - 135 kDa depending on cell type, binds ligands such as CD54 and CD169 (Siglec-1), and plays regulatory roles in cell adhesion and activation, apoptosis, and tumor progression. It is used as a marker for discriminating normal from neoplastic T and B cells within tissues, and binding of the CD43-10G7 clone to CD43 on neutrophils has been reported to induce homotypic cell adhesion. The antibody is purified by affinity chromatography and conjugated with PE under optimal conditions; the solution is free of unconjugated PE and unconjugated antibody.

The reagent is formulated in a phosphate-buffered solution at pH 7.2 containing 0.09% sodium azide and 0.2% (w/v) bovine serum albumin. For flow cytometry, the suggested use is 5 μL per 1 x 10^6 cells in 100 μL volume or per 100 μL of whole blood, with titration recommended for optimal performance. The product should be stored undiluted at 2 - 8°C, protected from prolonged exposure to light, and not frozen. The safety data sheet describes the material as a clear liquid that is not classified as hazardous under the Globally Harmonized System or the 2012 OSHA Hazard Communication Standard (29 CFR 1910.1200).
