Description

A mouse monoclonal antibody from STEMCELL Technologies (clone HIP8) reacts with the ~125 kDa GPIIbα subunit of human CD41, a heterodimeric integrin expressed primarily on platelets, megakaryocytes, monocytes, and acute megakaryoblastic leukemia cells. The GPIIbα subunit is linked by a single disulfide bond to a smaller ~25 kDa subunit (GPIIbβ). CD41 associates non-covalently with CD61 (integrin β3) in a calcium-dependent manner to form the CD41/CD61 (GPIIb/IIIa) complex, identified as a receptor for ligands including fibrinogen, fibronectin, vitronectin, and thrombin. Binding involves an RGD tripeptidyl sequence in the ligand.

The HIP8 antibody reportedly blocks platelet aggregation and inhibits platelet activation by several compounds. The reagent is supplied as a PE-conjugated preparation in clear liquid form. According to the manufacturer's Safety Data Sheet, the product is classified as a laboratory reagent that is not considered hazardous by the 2012 OSHA Hazard Communication Standard (29 CFR 1910.1200). It is intended for research use only and not for human or animal diagnostic or therapeutic applications unless otherwise stated; the manufacturer's quality management system is certified to ISO 13485. The supplier, STEMCELL Technologies Canada Inc., is based in Vancouver, British Columbia.

The antibody is of mouse IgG1, kappa isotype and targets the CD41 antigen (Gene ID 3674). Reported applications include flow cytometry, CyTOF, functional assays, immunofluorescence, immunohistochemistry, and neutralization/blocking. The antibody has been reported to react with baboon, capuchin monkey, chimpanzee, cynomolgus monkey, human, pig, and rhesus samples.
