Description

From STEMCELL Technologies, this APC-conjugated mouse monoclonal IgG1, kappa antibody (Clone HIP8) targets the ~125 kDa GPIIbα subunit of human CD41, a heterodimeric member of the integrin family expressed primarily on platelets, megakaryocytes, monocytes, and acute megakaryoblastic leukemia cells. The antibody has been reported to block platelet aggregation and to inhibit platelet activation by ADP, epinephrine, and collagen. It recognizes a ligand-binding complex: the GPIIbα subunit pairs via a single disulfide bond with a ~25 kDa GPIIbβ subunit and associates non-covalently with CD61 (integrin β3) in a calcium-dependent manner to form the CD41/CD61 (GPIIb/IIIa) receptor for fibrinogen, fibronectin, von Willebrand factor, vitronectin, plasminogen, thrombin, and thrombospondin.

Alternative designations for the target antigen include GPIIb, Integrin alpha 2b, Integrin alpha llb, ITGA2B, and Platelet glycoprotein IIb (Gene ID 3674). The antibody was raised in BALB/c mice against purified platelet membrane glycoproteins and purified by affinity chromatography before being conjugated with APC under optimal conditions. Reactivity has been reported in human, rhesus, cynomolgus, baboon, capuchin monkey, chimpanzee, and pig, and flow cytometry is the verified application; the antibody has also been validated for cell purity assessments following isolation with EasySep™ and RosetteSep™ kits.

For flow cytometry, the manufacturer recommends ≤ 5 μL per 1 x 10^6 cells in 100 μL, or per 100 μL whole blood, with titration advised for each application. The product is supplied in phosphate-buffered solution (pH 7.2) containing 0.09% sodium azide and 0.2% (w/v) bovine serum albumin, and should be stored at 2 - 8°C undiluted, protected from prolonged exposure to light and not frozen.
