Anti-Hu CD4, SK3, PE, 100tests

A mouse monoclonal IgG1, kappa antibody against human CD4 is offered by STEMCELL Technologies. The SK3 clone, raised in BALB/c mice against human peripheral blood T lymphocytes, is conjugated to PE (phycoerythrin). CD4 (alternative names Leu3, T4; Gene ID 920) is an approximately 59 kDa single-chain type I transmembrane glycoprotein belonging to the immunoglobulin superfamily, with four extracellular Ig-like domains (D1-D4); the SK3 epitope maps to the D3 domain. CD4 is expressed at relatively high levels on most thymocytes and on the T helper cell subpopulation, and at lower levels on peripheral blood monocytes and macrophages.

The antibody reacts with human, rhesus, cynomolgus, African green monkey, olive baboon, pigtailed macaque, sooty mangabey, and squirrel monkey CD4. It is verified for flow cytometry (FC), with a suggested use of 5 µL per 1 x 10^6 cells in 100 µL or per 100 µL of whole blood; titration is recommended for optimal performance. CD4 functions as a co-receptor that enhances the affinity of the TCR for MHC II-antigen complexes, and SK3 binding has been reported to block HIV attachment and the mixed lymphocyte reaction (MLR).

The reagent is supplied as a clear liquid in phosphate-buffered solution, pH 7.2, containing 0.09% sodium azide and 0.2% (w/v) bovine serum albumin, and is free of unconjugated PE and unconjugated antibody following purification by affinity chromatography. Store undiluted at 2 - 8°C; do not freeze, and protect from prolonged exposure to light. The product is not classified as hazardous under GHS and is intended for research use only.
