Description

From STEMCELL Technologies, the Anti-Human CD4 antibody (clone OKT4) targets CD4, an approximately 59 kDa single-chain type 1 transmembrane glycoprotein belonging to the Ig superfamily. CD4 contains four extracellular Ig-like domains (D1–D4), and the OKT4 epitope has been mapped to the D3 domain.

CD4 is expressed at relatively high levels on most thymocytes and on T-helper cells, and at lower levels on peripheral blood monocytes and macrophages. It binds a non-polymorphic region of MHC class II and acts as a co-receptor for the T cell receptor (TCR) in MHC II-restricted antigen recognition, enhancing TCR–MHC II–antigen affinity. CD4 also amplifies TCR signaling through its intracellular domain via cytoplasmic tyrosine kinases such as Lck and serves as a receptor for HIV, although binding of OKT4 does not block HIV attachment. The antibody has been verified for purity assessments of cells isolated with EasySep kits.

This is a biotin-conjugated mouse IgG2b, kappa primary antibody suitable for flow cytometry, functional assays, immunofluorescence, immunohistochemistry, and immunoprecipitation on cell types including T cells, CD4+ T cells, and regulatory T cells. It has been validated in chimpanzee, cynomolgus monkey, human, rhesus monkey, and DeBrazza's monkey. Additional conjugations offered across the product family include Alexa Fluor 488, APC, FITC, PE, PerCP-Cyanine5.5, and unconjugated formats. Relevant research areas include immunology and transplantation research.
