Description

STEMCELL Technologies offers this mouse monoclonal antibody specific for human CD34, a type 1 transmembrane glycoprotein of approximately 105 - 120 kDa. The antibody, clone 581 (isotype IgG1, kappa), was raised against human CD34+ leukemic cells and is conjugated to FITC (fluorescein isothiocyanate), with the solution free of unconjugated FITC following affinity chromatography purification.

CD34 is expressed on the surface of most human hematopoietic stem and progenitor cells (HSPCs), as well as on mesenchymal stem cells, embryonic fibroblasts, endothelial cells, neurons, and some tumor cell lines. It is used as a marker to identify and isolate HSPCs capable of cell engraftment. CD34 is expressed only transiently during hematopoiesis, so the frequency of CD34+ cells is low in bone marrow or cord blood (~1 - 5%) and very low (~0.1 - 0.5%) in peripheral blood. The 581 clone recognizes a class III CD34 epitope that is resistant to cleavage by neuraminidase and O-glycoprotease.

Clone 581 has been verified for flow cytometry (FC). It is recommended for purity assessments of cells isolated with the EasySep™ Human CD34 Positive Selection Kit and for labeling human mesenchymal cells cultured in the MesenCult™ Proliferation Kit. The reagent is a clear liquid formulated in a phosphate-buffered solution at pH 7.2, containing 0.09% sodium azide and 0.2% (w/v) bovine serum albumin. For flow cytometry, the suggested use is 5 μL per 1 x 10^6 cells in 100 μL, or per 100 μL of whole blood. The product is stable at 2 - 8°C when stored undiluted; it should not be frozen and should be protected from prolonged exposure to light. According to the supplier's safety data sheet, the preparation is not classified as a hazardous substance or mixture according to the Globally Harmonized System (GHS).
