Description

A biotin-conjugated mouse monoclonal antibody from STEMCELL Technologies, Clone 581 binds to human CD34, an ~105 - 120 kDa type 1 transmembrane glycoprotein expressed on the surface of most human hematopoietic stem and progenitor cells (HSPCs), as well as on mesenchymal stem cells, embryonic fibroblasts, endothelial cells, neurons, and some tumor cell lines. CD34 is expressed only transiently during hematopoiesis, with a frequency of CD34+ cells of ~1 - 5% in bone marrow or cord blood and ~0.1 - 0.5% in peripheral blood. The antigen serves as a marker for identifying and isolating HSPCs capable of cell engraftment and is thought to mediate attachment of stem cells to the bone marrow extracellular matrix or directly to stromal cells during early hematopoiesis, while also being involved in lymphocyte recruitment through binding to L- and E-selectin.

The 581 antibody recognizes a class III CD34 epitope that is resistant to neuraminidase and O-glycoprotease cleavage. It is a mouse IgG1, kappa antibody raised against human CD34+ leukemic cells. Verified for flow cytometry, this clone has also been verified for purity assessments of cells isolated with EasySep™ kits, including the EasySep™ Human CD34 Positive Selection Kit and the EasySep™ Human Whole Blood CD34 Positive Selection Kit, as well as for labeling human mesenchymal cells grown in MesenCult™ Proliferation Kit (Human). For flow cytometry, the suggested use is ≤ 1.0 µg per 1 x 10e6 cells in 100 µL volume. Clone 581 is verified for cell isolation and flow cytometry.

Provided as a phosphate-buffered solution at pH 7.2 containing 0.09% sodium azide, the antibody is purified by affinity chromatography and conjugated with biotin under optimal conditions, and is free of unconjugated biotin. The product is stable at 2 - 8°C when stored undiluted; it should not be frozen and should be protected from prolonged exposure to light.
