Description

STEMCELL Technologies offers a mouse monoclonal antibody (Clone 581) specific for human CD34 and conjugated to the fluorescent dye allophycocyanin (APC). CD34 is an approximately 105 - 120 kDa type 1 transmembrane glycoprotein expressed on the surface of most human hematopoietic stem and progenitor cells (HSPCs), as well as on mesenchymal stem cells, embryonic fibroblasts, endothelial cells, neurons, and some tumor cell lines. The marker is widely used to identify and isolate HSPCs capable of cell engraftment, and is thought to mediate attachment of stem cells to the bone marrow extracellular matrix or to stromal cells during early hematopoiesis, as well as lymphocyte recruitment through binding to L- and E-selectin. Clone 581 recognizes a class III epitope that is resistant to neuraminidase and O-glycoprotease cleavage.

The antibody is of the IgG1, kappa isotype and was raised against human CD34+ leukemic cells, then purified by affinity chromatography and conjugated with APC. It is supplied as a clear liquid in a phosphate-buffered solution at pH 7.2 containing 0.09% sodium azide and 0.2% (w/v) bovine serum albumin, and is free of unconjugated APC and unconjugated antibody. The reagent has been verified for flow cytometry, for purity assessments of cells isolated with the EasySep™ Human CD34 Positive Selection Kit, and for labeling human mesenchymal cells grown in MesenCult™ Proliferation Kit (Human). For flow cytometry, the manufacturer's suggested use is 5 μL per 1 x 10^6 cells in 100 μL, or per 100 μL of whole blood, with titration recommended for optimal performance.

Store the product undiluted at 2 - 8°C; do not freeze and protect from prolonged exposure to light. According to the manufacturer's Safety Data Sheet, the product is not classified as a hazardous substance or mixture under the 2012 OSHA Hazard Communication Standard (29 CFR 1910.1200) or the Globally Harmonized System (GHS). The supplier is STEMCELL Technologies Canada Inc., based in Vancouver, British Columbia, Canada.
