Description

A mouse monoclonal IgG1, kappa antibody from STEMCELL Technologies, clone HIM3-4, conjugated with PE (phycoerythrin) and reactive with human and chimpanzee CD33, also referred to as Siglec-3, gp67, p67, or SIGLEC3. CD33 is an ~67–75 kDa type I transmembrane sialic acid–binding immunoglobulin-like lectin (sialoadhesion protein) of the immunoglobulin superfamily. The antibody was purified by affinity chromatography and supplied in a phosphate-buffered solution at pH 7.2 containing 0.09% sodium azide, 0.1% gelatin, and 0.2% (w/v) bovine serum albumin. It is verified for flow cytometry and reported for FACS applications.

CD33 is expressed on committed myeloid progenitor cells, monocytes, granulocytes, dendritic cells, mast cells, resident macrophages, subsets of B cells, and activated T and NK cells, and serves as a major marker for differentiating myeloid from lymphoid or erythroid leukemias. In peripheral blood it is present at high levels on monocytes and at lower levels on granulocytes. Clone HIM3-4 binds to the extracellular C2 Ig-like domain of CD33 and recognizes both identified isoforms, CD33M and CD33m; it does not block subsequent binding of anti-CD33 clones P67.6, WM53, or WM54.

For flow cytometry, the suggested use is ≤5 µL (0.5 µg) per 1 × 10⁶ cells in 100 µL, with titration recommended for optimal performance for each application. The product is stable at 2–8 °C when stored undiluted; it should not be frozen and should be protected from prolonged exposure to light. According to the manufacturer's safety data sheet, it is not classified as hazardous under the GHS or the 2012 OSHA Hazard Communication Standard (29 CFR 1910.1200). The product is for research use only and is not intended for human or animal diagnostic or therapeutic uses unless otherwise stated.
