Description

A FITC-conjugated mouse monoclonal IgG1, kappa reagent from STEMCELL Technologies, clone 5E11.3.1, is raised against human epithelial cells (CD326/EpCAM) using the human mammary carcinoma cell line T47D as immunogen. The antibody was purified by affinity chromatography and is supplied in a formulation of phosphate-buffered saline containing 0.1% bovine serum albumin and < 0.1% sodium azide.

The 5E11.3.1 clone reacts with a surface marker expressed on most luminal cells of normal human mammary epithelium, with weak or absent labeling within the basal cell compartment and variable labeling on malignant breast epithelium. Positive labeling has been reported on the MCF 7, CAMA, T47D, BT-10, BT-20, COLO 205, and WiDr cell lines, while labeling is negative on human bone marrow cells, blood cells, lymphoid and mesenchymal tissues, mammary and marrow fibroblast cultures, and the human embryonic stem cell line H9. According to the manufacturer, CD326 (EpCAM) is expressed on the basolateral surfaces of epithelial cells.

Verified for flow cytometry and reported for FACS, the clone has been used for cell isolation, including purity assessments of cells isolated with EasySep™ kits. For flow cytometry, the suggested use is 20 µL per 1 × 10^6 cells in 100 µL; the antibody also works on paraffin sections when a heat-induced antigen retrieval protocol is applied. The product is a clear liquid and is stable at 2 - 8 °C when stored undiluted; it should not be frozen and should be protected from prolonged exposure to light. The manufacturer's safety data sheet states that the product is not classified as a hazardous substance or mixture according to GHS, and notes it is harmful to aquatic life with long lasting effects.
