Anti-Hu CD32, IV.3, FI, 100tests

This FITC-conjugated monoclonal antibody from STEMCELL Technologies, clone IV.3, targets human CD32 (FcγRII), an approximately 40 kDa type 1 transmembrane glycoprotein encoded by three genes (A, B, C) that generates at least six isoforms (IIa1, IIa2, IIb1, IIb2, IIb3 and IIc) through alternative mRNA splicing. Produced in mouse as IgG2b, kappa, the antibody was raised against the K-562 human erythromyeloblastoid leukemia cell line.

CD32 is expressed by monocytes/macrophages, placental trophoblasts and endothelial cells across all isoforms, with the IIb isoform additionally expressed by B cells, IIa by platelets, granulocytes and weakly by B cells, and IIc by NK cells and neutrophils. The IV.3 clone binds most strongly to the IIa isoforms of CD32, with its epitope mapped to amino acids 132-137 [FSHLDP] in domain 2, within the ligand binding site. CD32 binds weakly to the Fc region of monomeric IgG but more strongly to IgG aggregates and immune complexes, and the IV.3 antibody can be employed as a blocking antibody to reduce non-specific binding in antibody-based detection and cell separation experiments.

The antibody has been verified for flow cytometry and cell separation, including purity assessments of cells isolated with EasySep kits such as the EasySep Human T Cell Enrichment Kit and EasySep Human CD4+ T Cell Enrichment Kit. It is supplied in phosphate-buffered saline containing less than 0.1% sodium azide and less than 0.1% bovine serum albumin, purified by affinity chromatography and conjugated with FITC. The product should be stored undiluted at 2-8°C, protected from light, and not frozen.
