Description

This PE-conjugated mouse monoclonal antibody from STEMCELL Technologies, clone LNH-94, recognises human CD298 (ATP1B3), the β3 subunit of the Na+/K+ ATPase. The Na+/K+ ATPase is described as a 42 kDa type II transmembrane protein composed of four catalytic α subunits and three smaller glycosylated non-catalytic β subunits, and is important for establishing electrochemical gradients and maintaining normal resting membrane potential by coupling ATP hydrolysis to the active transport of Na+ and K+. The β subunits have been reported to act as chaperones that stabilise the α subunits and occlude K+.

The antibody is of the mouse IgG1, kappa isotype and was purified by affinity chromatography before conjugation with PE under optimal conditions; the resulting solution is free of unconjugated PE and unconjugated antibody. It is formulated in a phosphate-buffered solution, pH 7.2, containing 0.09% sodium azide and 0.2% (w/v) BSA. The verified application is flow cytometry, and for this application the suggested use is 5 μL per 1 x 10^6 cells in a 100 μL volume, with titration recommended for optimal performance in each application.

CD298 has been used to identify metastatic human cancer cells in mouse xenograft models and has been reported to modulate aspects of immunity, including HIV production and T cell and B cell activation. The antigen is found on leukocytes, platelets, and most other tissue types. The product is stable at 2 - 8 °C when stored undiluted; it should not be frozen and should be protected from prolonged exposure to light.
