Anti-Hu CD25, BC96, FI, 100tests

A FITC-conjugated mouse monoclonal IgG1, kappa antibody (clone BC96) from STEMCELL Technologies, raised against CD25 protein of human origin and purified by affinity chromatography. The reagent is supplied in a phosphate-buffered solution at pH 7.2 containing 0.09% sodium azide and 0.2% (w/v) BSA, conjugated with FITC, and free of unconjugated FITC. Verified and reported applications are flow cytometry (FC). The manufacturer's suggested use is 5 μL per 1 x 10^6 cells in 100 μL volume, with titration recommended for optimal performance.

The antibody recognizes CD25, a 55 kDa type I integral transmembrane glycoprotein also referred to as IL-2 Receptor alpha chain, IL-2Ra, IL2Ra, Tac, or p55. CD25 acts as a low-affinity receptor for Interleukin-2 (IL-2) and associates with IL-2 receptor β (CD122) and common γ (CD132) chains to form a high-affinity IL-2 receptor. It is described as present on activated T and B cells, monocytes, macrophages, and regulatory T cells, and as involved in the activation and proliferation of lymphocytes as well as in the identification of T cell subsets in viral infections such as HIV and HTLV. Co-expression of CD25 with CD4 and FOXP3 is described as a phenotypic signature for regulatory T cells. Listed species reactivity includes Human, Rhesus, Cynomolgus, Baboon, Chimpanzee, and Pig-tailed Macaque.

According to STEMCELL Technologies, the antibody has been verified for analyzing CD25 antigen expression on cultured human T cells activated with ImmunoCult Human CD3/CD28 T Cell Activator and ImmunoCult Human CD3/CD28/CD2 T Cell Activator. The product is stable at 2 - 8 °C when stored undiluted, should not be frozen, and should be protected from prolonged exposure to light. Representative flow cytometry data shown by the manufacturer used PHA-L-activated human peripheral blood mononuclear cells (PBMCs) labeled with this antibody alongside a Mouse IgG1, kappa Isotype Control Antibody, clone MOPC-21.
