Description

The Anti-Human CD22 Antibody, Clone HIB22 from STEMCELL Technologies is a mouse monoclonal IgG1, kappa antibody that recognizes CD22 (Siglec-2), a type I transmembrane glycoprotein expressed in the cytoplasm of pro-B and pre-B cells as well as on the surface of mature and activated B cells and B cell lymphomas. CD22 is not expressed on plasma cells.

CD22 functions as an inhibitory receptor for B cell receptor signaling and plays a role in modulating the activation threshold of B cells and controlling CD40 signaling. The molecule is a member of the sialoadhesion subgroup within the immunoglobulin superfamily, comprising an N-terminal V-type Ig domain followed by six C2-type Ig domains. Two isoforms are generated by alternative splicing: CD22α at ~130 kDa and CD22β at ~140 kDa. Monomeric CD22β is the most abundant isoform, with an αβ heterodimer also expressed. Binding of the HIB22 antibody reportedly requires the presence of the two N-terminal-most domains, recognizes both the cytoplasmic and surface-bound forms of CD22, and does not inhibit CD22-mediated cyto-adhesion.

The antibody is offered in unconjugated, APC, FITC, and PE formats and has been validated for flow cytometry applications. It is also cited for use in ELISA, functional assays, immunocytochemistry, immunofluorescence, immunohistochemistry, and immunoprecipitation, and reacts with human and chimpanzee samples. In flow cytometry data provided by the manufacturer, Clone HIB22 was applied to human peripheral blood mononuclear cells or buffy coat nucleated cells processed with the EasySep™ HLA Whole Blood B Cell Positive Selection Kit and the EasySep™ Human CD19 Positive Selection Kit, with conjugate-specific staining shown alongside matched mouse IgG1, kappa isotype controls. Gene ID: 933.
