Anti-Hu CD22, HIB22, PE, 100tests

This PE-conjugated monoclonal antibody from STEMCELL Technologies, clone HIB22, recognizes human and chimpanzee CD22 (Siglec-2), a type I transmembrane glycoprotein of the sialoadhesion subgroup within the immunoglobulin superfamily. The reagent is a mouse IgG1, kappa antibody purified by affinity chromatography and conjugated with PE under optimal conditions.

CD22 is expressed in the cytoplasm of pro-B and pre-B cells and on the surface of mature and activated B cells and B cell lymphomas, but not on plasma cells. It functions as an inhibitory receptor for B cell receptor signaling, modulating B-cell activation thresholds and CD40 signaling. Two isoforms arise by alternative splicing: CD22α (~130 kDa) and CD22β (~140 kDa); a CD22αβ heterodimer is also expressed. The HIB22 antibody reportedly requires the two N-terminal-most domains of CD22 for binding, and recognizes both cytoplasmic and surface-bound forms without inhibiting CD22-mediated cyto-adhesion. The antibody was raised against hairy cell leukemia cells, has been verified for flow cytometry (FC), and is reported for FC. It has been used to assess the purity of cells isolated with EasySep™ kits, including the HLA Whole Blood B Cell Positive Selection Kit and the Human CD19 Positive Selection Kit.

The product is supplied as a clear liquid in phosphate-buffered solution, pH 7.2, containing 0.09% sodium azide and 0.2% (w/v) bovine serum albumin, and is free of unconjugated PE and unconjugated antibody. STEMCELL Technologies recommends storage at 2 - 8 °C; the product should not be frozen and should be protected from prolonged exposure to light. For flow cytometry, the suggested use is 5 μL per 1 × 10^6 cells in 100 μL volume or per 100 μL of whole blood, with titration recommended for optimal performance. The product is classified as not a hazardous substance or mixture according to the Globally Harmonized System (GHS) and is for research use only.
