Description

From STEMCELL Technologies, this monoclonal antibody (clone HIB22) is specific for human CD22 and is conjugated to FITC. It is supplied as a laboratory reagent, with flow cytometry named as the primary intended application, and is also applicable to ELISA, functional assays, immunocytochemistry, immunofluorescence, immunohistochemistry, and immunoprecipitation. The antibody is of mouse origin, isotype IgG1, kappa, and shows reactivity against chimpanzee and human samples.

The HIB22 clone reacts with CD22 (Siglec-2), a type I transmembrane glycoprotein expressed in the cytoplasm of pro-B and pre-B cells and on the surface of mature and activated B cells and B cell lymphomas, but not on plasma cells. CD22 functions as an inhibitory receptor for B cell receptor signaling, modulating the activation threshold of B cells and controlling CD40 signaling. It comprises two isoforms generated by alternative splicing: CD22α (~130 kDa) and CD22β (~140 kDa), with monomeric CD22β being the most abundant isoform but an αβ heterodimer is also expressed. CD22 is a member of the sialoadhesion subgroup within the immunoglobulin superfamily, with an N-terminal V-type Ig domain followed by six C2-type Ig domains. HIB22 binding reportedly requires the presence of the two N-terminal-most domains; the antibody recognizes both cytoplasmic and surface-bound forms of CD22 and does not inhibit CD22-mediated cyto-adhesion.

The product is supplied as a clear liquid and is not classified as hazardous under the 2012 OSHA Hazard Communication Standard (29 CFR 1910.1200). Per supplier guidance it should be stored at 2–8 °C, protected from light, and not subjected to repeated freeze–thaw cycles. The product is not regulated for transport by DOT, TDG, MEX, ICAO, IATA, IMDG, RID, ADR, or ADN.
